Are you new to League of Legends?
Lvl 6 Mastery- to unlock lvl 6 mastery your champion must be in lvl 5 mastery and had 2 mastery tokens
Lvl7 Mastery- Champion lvl 6 and 3 tokens mastery

How to get mastery tokens? you need to earn "S-" and above for Lvl 6 mastery token and "S" and above for Lvl7 mastery token

How to unlok lvl maestry?

You have to perform well with your champion to earn mastery points. A quick google of "League of legends champion mastery points" should give you all the info you need
