Wow. Great question. Many mention the shooting of Archduke Ferdinand and that would have to be up there.
For the US it would be the assassination of Lincoln. So much changed as a result of that. With Lincoln there would have been reconciliation rather than harsh Reconstruction. Blacks would have realized their rights long before they actually did. With reconciliation there would have been no Jim Crow laws, no lynchings, no discrimination, perhaps no welfare state.
When Lincoln died, the future of the US was changed dramatically and for the worse. His death was the worse thing that could have happened to the North, the South, and more implying African Americans.
Every lost. No one won.
